Nani grabbed the winner when Orlando City defeated David Beckham's Inter Miami by 2-1 as the Major League Soccer (MLS) returned since its closure in March.
Before kickoff players took to the pitch and raised their fist for eight minutes and 46 seconds indicating how George Floyd was pinned to the floor by policemen.
It was the first game since the global pandemic and Nani's 97th-minute goal made the difference in the match after the lengthy break.
"It was emotional for the ones who were there," said Orlando's Nani.
"We all want to change the world. We want a better world - no differences, no discrimination. Everyone in the world should stop for a couple of minutes and think about our children and teach them how to be a better person and create a better world."
Players of both sides knelt on one leg around the center-circle as footballers from the other teams stood around them and one-by-one raised their fist.
They wore face masks with 'Black Lives Matter', written on each of them and also had T-shirts with slogans including 'silence is violence' and 'black and proud'.
It was the first match in the MLS returns Tournament which takes a group stage and knockout format and will end on August 11, 2020.
Juan Agudelo had put Orlando into the lead after the restart but Chris Mueller equalized for Inter in the 70th minute before Nani scored in time added on.
